Palvella Therapeutics, Inc. (Nasdaq: PVLA) announced on October 28, 2026, that the U.S. Food and Drug Administration (FDA) has accepted for filing and granted Priority Review to the company’s New Drug Application (NDA) for QTORIN™ 3.9% rapamycin anhydrous gel. The application targets the treatment of microcystic lymphatic malformations, a rare genetic disease affecting an estimated 30,000 or more people in the United States.
The FDA has assigned a Prescription Drug User Fee Act (PDUFA) target action date of February 28, 2027. This designation accelerates the review timeline, reflecting the potential significance of the therapy for patients with no currently approved treatments.
The NDA is supported by data from the Phase 3 SELVA study, which achieved statistical significance across all six prespecified efficacy endpoints (all p<0.001). The study reported that 95% of participants aged 6 years and older who completed the efficacy evaluation demonstrated improvement on the primary endpoint. QTORIN™ rapamycin has received Breakthrough Therapy, Fast Track, and Orphan Drug designations from the FDA.
Regarding safety, the press release notes that QTORIN™ rapamycin was well tolerated in the SELVA study, with no drug-related serious adverse events reported and systemic rapamycin concentrations remaining below 2 ng/mL at all measured timepoints.
Palvella stated that it is preparing for a potential standalone U.S. commercial launch in the first half of 2027, subject to FDA approval. The company has assembled a team including experienced rare disease commercial leadership, a field-based medical science liaison team, and a dedicated patient services organization to facilitate market access and patient support.
Beyond microcystic lymphatic malformations, Palvella is advancing QTORIN™ rapamycin for cutaneous venous malformations and clinically significant angiokeratomas, as well as QTORIN™ pitavastatin for disseminated superficial actinic porokeratosis.
