Coherus Oncology, Inc. announced interim clinical data from its TREGCHECK development program on October 28, 2026. The data evaluates tagmokitug, an investigational cytolytic CCR8 antibody, in combination with toripalimab, with or without chemotherapy.

The company reported encouraging anti-tumor activity across multiple solid tumors. In evaluable patients with previously untreated advanced or metastatic esophageal squamous cell carcinoma (ESCC), tagmokitug combined with toripalimab and chemotherapy demonstrated a confirmed overall response rate (cORR) of 75% irrespective of PD-L1 status. In PD-L1 positive patients, the cORR was 100% as of October 14, 2026. Subsequent to the data cutoff, two additional partial responses were confirmed, increasing the cORR to 89%.

In second-line ESCC patients who had failed prior chemotherapy, the 700 mg dose arm of tagmokitug combined with toripalimab showed a median progression-free survival of 5.5 months. This compares to an estimated standard of care benchmark of approximately 3.4 months. The company also reported clinical benefit rates and tumor shrinkage in other indications, including head and neck and gastrointestinal tumors.

Safety data from more than 120 patients treated with tagmokitug plus toripalimab indicated a manageable profile. The most common treatment-related adverse events were infusion-related reactions (25.0%), fatigue (19.2%), and rash (15.8%). No Grade 5 treatment-related adverse events were reported.

Based on these findings, Coherus plans to expand enrollment in first-line and second-line ESCC cohorts and continue evaluating the durability of responses and biomarker findings. The company also noted it is advancing tagmokitug in combinations beyond PD-1 inhibition, including a planned study evaluating safety with a T-cell engager in metastatic castration-resistant prostate cancer.
